چکیده

Several papers have been published on the biosynthesis of glucosamine in different organisms (l-3), but the reactions which lead to the synthesis of galactosamine have hardly been explored. The first indication of its mode of synthesis came from the isolation of uridine diphosphate’ acetylgalactosamine from liver by Pontis (4). This finding showed that the transformation of acetylhexosamines is similar to that of the pair glucosegalactose which has been studied in detail (5, 6). Pontis mentioned that, upon incubation of UDP-acetylglucosamine with a crude liver extract, some acetylgalactosamine was formed, but he did not investigate whether the substance appeared in the free or combined form. Further work on the point is reported in this paper. The products arising by the action of rat liver homogenate on UDP-acetylglucosamine were found to be free acetylgalactosamine and uridine monophosphate.

Enzymatic Sulfation of Mucopolysaccharides in Hen Oviduct

An enzymatic reaction has been identified in the isthmus of hen oviduct in which sulfate is transferred from 3’-phosphoadenosine 5’-phosphosulfate to mucopolysaccharide acceptors (1). From studies of the reaction with monosaccharides and oligosaccharides as model acceptors (‘2) it was concluded that one or more enzymes in the preparations employed catalyze a direct transfer of sulfate from PAPS...

Enzymatic synthesis of the protein-hexosamine linkage in sheep submaxillary mucin.

A particulate enzyme preparation, obtained from sheep submaxillary gland, catalyzed the transfer of iV-acetyl-Dgalactosamine from its uridine diphosphate derivative to certain protein acceptors. The proteins were prepared from sheep submaxillary mucin by enzymatic or chemical treatments that removed the disaccharide residues from the mucin. Only these proteins served as N-acetylgalactosamine ac...

The enzymatic desulfation of sulfated sugar nucleotides by a novel sulfatase in hen oviduct.

A soluble fraction from the isthmus region of hen oviduct catalyzes the liberation of SOF from UDP-N-acetylgalactosamine-4-sulfate (the nucleotide that accumulates in the same tissue during egg formation). The enzyme is purified some &fold and is shown to catalyze the reactions: (a) UDP-N-acetylgalactosamine-4-sulfate 4 UDP-N-acetylgalactosamine plus SOF; and (b) UDP-N-acetylgalactosamine-4,6-d...

Enzymatic Transfer of Sulfate from 3’-

The enzymes catalyzing the sulfation by 3’-phosphoadenosine 5’-phosphosulfate of chondroitin, chondroitin sulfates A, B, and C, and heparitin sulfate have been reported by Suzuki and Strominger (1) and Suzuki, Threnn, and Strominger (2) to occur in the isthmus region of hen oviduct’. The enzyme preparations used in these studies showed an absolute dependence on addition of the mucopolysaccharid...

Ganglioside biosynthesis in mouse embryos: sialyltransferase IV and the asialo pathway.

The in vitro activity of sialyltransferase IV (SAT-IV), which catalyzes the transfer of sialic acid to the terminal galactose of different gangliotetraosylceramides (GA1, GM1a and GD1b), was examined in membrane-enriched preparations from mouse embryos at embryonic day 12 (E-12). Gangliosides GD1a and GT1b were the only reaction products using GM1a and GD1b as substrates, respectively. The Km v...
